Технология тканевых матриц (Tissue Microarray, TMA) и ее применение в иммуногистохимии.
TMA Master и TMA Grand Master - системы получения тканевых матриц
из парафиновых блоков для решения диагностических и исследовательских задач,
проведения клинических испытаний. С помощью приборов для подготовки ТМА матриц
формируются реципиентные блоки, которые включают в себя большое количество зон
интереса с различных донорских блоков.
Для оценки качества коллекции множественых микрообразцов тканей на парафиновом блоке, каждый десятый срез рекомендуется окрашивать рутинной окраской.
Мастер блок (tissue microarray, тканевая матрица, множественно-тканный парафиновый блок, тканевой микрочип) - это парафиновый блок, в который множественные столбики ткани перемещены из обычных парафиновых(донорских) блоков в организованном порядке. В соответствии с дизайном, диаметр столбиков может быть от 0,6 до 2,0 мм. Высота столбиков также варьирует.
В дальнейшем парафиновый блок нарезается на микротоме. Срезы толщиной от 3 до 5 микрон помещаются на стекла, которые затем могут быть использованы в различных экспериментах.
Эксперименты включают: гистохимическое, иммуногистохимичекое, иммунофлуоресцентное исследования, гибридизацию in situ, и даже техники микродиссекции.

Вырезка – точка отсчета качества пробоподготовки гистологического препарата в
лаборатории. Только использование специального инструментария позволит
врачу-патологоанатому провести качественную и, что не менее важно,
стандартизованную процедуру вырезки.

В статье кратко описана сущность методики и результаты исследования представительной выборки клинико-морфологического материала.
Множественно-тканный парафиновый блок применяется для исследования материала от десятков и сотен пациентов с множественными маркерами одновременно и экономным использованием реагентов для ИГХ.

Recipient Block Indexer for MTA-1
Recipient block indexer, a rotating recipient block holder that holds four
blocks at a time and speeds up the concurrent production of multiple blocks.
Punches & Stylets (Consumables)
Punches are sold as matched pairs (one for the donor block and another for the
recipient block).
Which punch size to choose?
Standard size punches: 0.6 mm diameter
Advantages:
- less damage to the donor block
- more samples from any given donor
- more samples in any given recipient block
- easiest to remove the cores from the donor blocks
- easiest to remove the blank cores from the recipient block
Custom-size punches: 1.0, 1.5 and 2.0 mm diameter
Advantages:
- More robust, harder to damage (e.g., during training)
- larger individual sample size Potential Disadvantages:
- A tiny fraction of users have reported blocks broken by the larger
punches.
- A larger but still small fraction of users have reported difficulty in
removing cores from blocks when using the larger punches.
